NMN (β-Nicotinamide Mononucleotide) is the direct biochemical precursor to NAD+. It is converted to NAD+ inside cells by the enzyme NMNAT (nicotinamide mononucleotide adenylyltransferase). A dedicated NMN transporter, Slc12a8, has been identified in the small intestine and other tissues, enabling direct cellular uptake without prior dephosphorylation to NR. By replenishing the intracellular NMN pool, supplementation supports NAD+ biosynthesis via the salvage pathway — the primary route by which cells maintain NAD+ levels.
