ACTH 1-39 binds MC2R (melanocortin 2 receptor) on adrenocortical zona fasciculata cells, activating adenylyl cyclase and cAMP-PKA signalling. This stimulates steroidogenic acute regulatory protein (StAR) and cholesterol side-chain cleavage enzyme (CYP11A1) to produce cortisol from cholesterol.
